Summary
Dronedarone inhibits CYP3A4 and P-glycoprotein, both of which handle bosutinib. Exposure to bosutinib may rise, increasing gastrointestinal, hepatic and myelosuppressive toxicity.
What this interaction means in plain words
Bosutinib and Dronedarone can interact in a way that may be serious. Using them together is sometimes necessary, but it needs a deliberate decision by the prescriber, a plan for monitoring, and clear advice on the warning signs to watch for.
Bosutinib belongs to Oncology; Dronedarone belongs to Cardiovascular.
Why it happens
Dual inhibition of CYP3A4 metabolism and P-glycoprotein efflux of bosutinib.
What to do
Avoid concurrent use if possible and consider an alternative antiarrhythmic. If used together, monitor liver tests, blood counts and GI tolerability and consider reducing the bosutinib dose.
