Summary

Mitotane strongly induces CYP3A4 and can substantially lower bromocriptine concentrations. This may lead to loss of prolactin or growth hormone control or reduced antiparkinsonian effect, and the induction persists for weeks after mitotane is stopped.

What this interaction means in plain words

Bromocriptine and Mitotane can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.

Bromocriptine belongs to Central Nervous System; Mitotane belongs to Oncology.

Evidence level: Moderate

Why it happens

Mitotane induces CYP3A4, increasing the metabolic clearance of bromocriptine.

What to do

Monitor prolactin or clinical response and increase the bromocriptine dose if needed. Reduce the dose again after mitotane is discontinued.

What to ask your pharmacist

  • Should I take them at different times of day?
