Summary
Mitotane is a long-acting enzyme inducer and is expected to reduce doravirine levels substantially. The induction can persist for weeks after mitotane is stopped, creating a risk of virologic failure.
What this interaction means in plain words
Doravirine and Mitotane are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
Doravirine belongs to Anti Virals; Mitotane belongs to Oncology.
Why it happens
Mitotane induces CYP3A4, increasing doravirine metabolism.
What to do
Avoid coadministration and choose an antiretroviral regimen not dependent on CYP3A. Allow for an extended washout of mitotane before using doravirine.
