Summary
Chloramphenicol may increase dasatinib concentrations because dasatinib is a CYP3A4 substrate. Higher exposure may worsen myelosuppression, fluid retention and QT prolongation. Chloramphenicol itself also suppresses bone marrow, so hematologic toxicity may be additive.
What this interaction means in plain words
Chloramphenicol and Dasatinib can interact in a way that may be serious. Using them together is sometimes necessary, but it needs a deliberate decision by the prescriber, a plan for monitoring, and clear advice on the warning signs to watch for.
Chloramphenicol belongs to Anti Bacterial; Dasatinib belongs to Oncology.
Why it happens
Chloramphenicol inhibits CYP3A4-mediated dasatinib metabolism, and both drugs have myelosuppressive effects.
What to do
Avoid the combination if an alternative antibiotic exists. Otherwise monitor complete blood counts and ECG, and consider reducing the dasatinib dose.
What to ask your pharmacist
- Does the benefit of taking both outweigh the risk in my case?
- Do I need any blood test, ECG or other check while on both?
- What early warning signs should I watch for, and what should I do if they appear?
