Summary

Both drugs can prolong the QT interval, so giving them together may add to the risk of torsades de pointes and other ventricular arrhythmias. Risk is higher with electrolyte disturbances, bradycardia or existing cardiac disease.

What this interaction means in plain words

Chlorpromazine and Dabrafenib can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.

Chlorpromazine belongs to Central Nervous System; Dabrafenib belongs to Oncology.

Evidence level: Low

Why it happens

Additive QT prolongation through pharmacodynamic effects on cardiac repolarization; dabrafenib may also induce CYP3A4 and lower chlorpromazine exposure to a minor degree.

What to do

Obtain a baseline ECG and repeat it after starting or changing doses, and correct potassium and magnesium abnormalities. Consider an alternative if the QTc becomes markedly prolonged.

What to ask your pharmacist

  • Should I take them at different times of day?
  • Does the dose of either medicine need to change?
  • Is there anything I should avoid, such as alcohol or certain foods, while on both?
