Summary
Darunavir, usually boosted with ritonavir or cobicistat, strongly inhibits CYP3A4 and can greatly raise eletriptan exposure. This increases the risk of serious vasoconstrictive effects. Eletriptan labeling contraindicates use within 72 hours of potent CYP3A4 inhibitors.
What this interaction means in plain words
Darunavir and Eletriptan are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
Darunavir belongs to Anti Virals; Eletriptan belongs to Central Nervous System.
Why it happens
Potent inhibition of CYP3A4-mediated eletriptan metabolism, mainly via the ritonavir or cobicistat booster.
What to do
Do not give eletriptan within 72 hours of boosted darunavir. Use a triptan that is not metabolized by CYP3A4, such as sumatriptan or rizatriptan.
