Summary

Cyclosporine inhibits OATP1B1 and substantially increases elagolix exposure, raising the risk of adverse effects such as bone loss, liver enzyme elevation and mood changes. Elagolix labeling prohibits use with strong OATP1B1 inhibitors such as cyclosporine.

What this interaction means in plain words

Cyclosporin and Elagolix are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.

Cyclosporin belongs to Miscellaneous; Elagolix belongs to Musculoskeletal.

Evidence level: High

Why it happens

Cyclosporine inhibits OATP1B1-mediated hepatic uptake of elagolix, increasing its plasma concentration.

What to do

Do not coadminister; choose alternative endometriosis therapy while cyclosporine is needed.
