Summary
Boosted darunavir inhibits CYP3A4 and can raise ergotamine levels substantially. This can produce acute ergot toxicity with peripheral vasospasm and ischemia.
What this interaction means in plain words
Darunavir and Ergotamine are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
Darunavir belongs to Anti Virals; Ergotamine belongs to Central Nervous System.
Why it happens
Ritonavir or cobicistat-boosted darunavir inhibits CYP3A4, reducing ergotamine clearance.
What to do
Do not co-administer; use a non-ergot alternative for migraine.
