Summary
Darunavir, usually boosted with ritonavir or cobicistat, strongly inhibits CYP3A4 and may raise donepezil levels. This can increase cholinergic effects including bradycardia, syncope and gastrointestinal symptoms, and donepezil can also prolong the QT interval.
What this interaction means in plain words
Darunavir and Memantine-donepezil can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Darunavir belongs to Anti Virals; Memantine-donepezil belongs to Central Nervous System.
Why it happens
Boosted darunavir inhibits CYP3A4, reducing donepezil clearance.
What to do
Monitor heart rate, ECG if risk factors exist, and cholinergic adverse effects. Consider reducing the donepezil dose if these effects develop.
What to ask your pharmacist
- Should I take them at different times of day?
- Does the dose of either medicine need to change?
