Summary

Denosumab carries a risk of serious infections, and concurrent immunosuppressants such as prednisone may add to that risk. This includes skin, urinary tract, and other bacterial infections. Prednisone also causes bone loss, so the combination is often used intentionally for glucocorticoid-induced osteoporosis.

What this interaction means in plain words

Denosumab and Prednisone can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.

Denosumab belongs to Oncology; Prednisone belongs to Endocrine.

Evidence level: Moderate

Why it happens

Additive immunosuppression, with denosumab altering RANKL-mediated immune signaling and prednisone suppressing immune function broadly.

What to do

Counsel patients to report signs of infection early and monitor for infection during therapy. Ensure adequate calcium and vitamin D and monitor serum calcium.
