For healthcare professionals — reference only. This page is a brief summary, not medical advice and not a substitute for official labeling or clinical judgement. Always verify before prescribing or dispensing.

Summary

Gepirone is a serotonin 5-HT1A agonist antidepressant whose labeling contraindicates use with MAO inhibitors. Combining it with safinamide risks serotonin syndrome, with agitation, hyperthermia, neuromuscular hyperactivity and autonomic instability.

What this interaction means in plain words

Gepirone and Safinamide are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.

Gepirone belongs to Central Nervous System; Safinamide belongs to Central Nervous System.

Evidence level: Moderate

Why it happens

MAO inhibition elevates serotonin levels, adding to the serotonergic activity of gepirone.

What to do

Do not co-administer. Observe a washout of at least 14 days after stopping an MAOI before starting gepirone, and at least 14 days after stopping gepirone before starting an MAOI.

What to ask your pharmacist

  • Is there an alternative to one of these two medicines for me?
  • If both are essential, who is monitoring me and how often?
  • Which symptoms mean I should stop and seek help straight away?
