Summary
Leniolisib inhibits hepatic OATP1B1 and OATP1B3 uptake transporters, which may reduce hepatic uptake of statins and raise their systemic levels. Higher lovastatin exposure increases the risk of myopathy and rhabdomyolysis.
What this interaction means in plain words
Leniolisib and Lovastatin can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Leniolisib belongs to Miscellaneous; Lovastatin belongs to Cardiovascular.
Why it happens
Leniolisib inhibits OATP1B1/1B3-mediated hepatic uptake of lovastatin and its acid metabolite.
What to do
Use the lowest effective lovastatin dose, or consider a statin less dependent on OATP such as fluvastatin. Monitor for muscle symptoms and check CK if they occur.
What to ask your pharmacist
- Should I take them at different times of day?
- Does the dose of either medicine need to change?
- Is there anything I should avoid, such as alcohol or certain foods, while on both?
