Summary
Mifepristone inhibits CYP3A4 and can substantially raise lovastatin exposure, a drug that depends heavily on this enzyme for clearance. Higher lovastatin levels greatly increase the risk of myopathy and rhabdomyolysis.
What this interaction means in plain words
Lovastatin and Mifepristone are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
Lovastatin belongs to Cardiovascular; Mifepristone belongs to Endocrine.
Why it happens
Mifepristone inhibits CYP3A4-mediated first-pass and hepatic metabolism of lovastatin, increasing systemic exposure.
What to do
Do not use together; hold lovastatin or switch to a statin less dependent on CYP3A4, such as pravastatin or rosuvastatin, during mifepristone treatment.
