Summary
Tepotinib inhibits P-glycoprotein and may raise exposure to methotrexate, which is also a P-gp substrate. Higher methotrexate levels can increase myelosuppression, mucositis, hepatotoxicity and renal toxicity.
What this interaction means in plain words
Methotrexate and Tepotinib can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Methotrexate belongs to Oncology; Tepotinib belongs to Oncology.
Why it happens
Tepotinib inhibits P-gp efflux transport, reducing methotrexate elimination and increasing its systemic exposure.
What to do
Monitor blood counts, renal and liver function, and methotrexate levels where high-dose therapy is used. Reduce the methotrexate dose if signs of toxicity appear.
What to ask your pharmacist
