Summary
Nilotinib may inhibit CYP2D6, reducing conversion of tramadol to its active metabolite and altering the balance between opioid and monoaminergic effects. Both drugs can also prolong the QT interval or lower seizure threshold to some degree, so overall tolerability may be affected.
What this interaction means in plain words
Nilotinib and Tramadol can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Nilotinib belongs to Oncology; Tramadol belongs to Musculoskeletal.
Why it happens
Possible CYP2D6 inhibition by nilotinib reduces tramadol activation; nilotinib also has QT-prolonging effects.
What to do
Monitor analgesic response and ECG/QT in patients with other risk factors. Consider an alternative analgesic if QT risk is a concern.
What to ask your pharmacist
- Should I take them at different times of day?
- Does the dose of either medicine need to change?
- Is there anything I should avoid, such as alcohol or certain foods, while on both?
