Summary

Pentobarbital is a potent sedative-hypnotic and prochlorperazine adds further CNS depression and hypotension. Excess sedation and respiratory depression may occur. Barbiturates can also induce metabolism of phenothiazines and reduce their effect.

What this interaction means in plain words

Pentobarbital and Prochlorperazine can interact in a way that may be serious. Using them together is sometimes necessary, but it needs a deliberate decision by the prescriber, a plan for monitoring, and clear advice on the warning signs to watch for.

Evidence level: Moderate

Why it happens

Additive CNS depression; barbiturate induction of hepatic enzymes may lower prochlorperazine exposure.

What to do

Monitor sedation, respiratory status and blood pressure, and lower doses as needed. Reassess antiemetic or antipsychotic efficacy during prolonged barbiturate use.

What to ask your pharmacist

  • Does the benefit of taking both outweigh the risk in my case?
  • Do I need any blood test, ECG or other check while on both?
  • What early warning signs should I watch for, and what should I do if they appear?
