Summary
Temozolomide causes lymphopenia and myelosuppression, which can impair CAR T-cell expansion and add to infection risk after CAR T therapy. Chemotherapy given near the time of CAR T infusion, other than planned lymphodepletion, may compromise efficacy and safety.
What this interaction means in plain words
Temozolomide and Tisagenlecleucel can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Temozolomide belongs to Oncology; Tisagenlecleucel belongs to Oncology.
Why it happens
Additive myelosuppressive and lymphotoxic effects of temozolomide may reduce CAR T-cell persistence and increase immunosuppression-related infection.
What to do
Avoid temozolomide in the period around leukapheresis and CAR T infusion unless part of the planned lymphodepletion or bridging regimen. Monitor blood counts and infection signs if given afterward.
