Summary
Valbenazine is a VMAT2 inhibitor that depletes monoamines, and MAOIs raise synaptic monoamine levels. The combination may reduce valbenazine efficacy and may increase the risk of serotonin toxicity. Labeling contraindicates concomitant use with MAOIs.
What this interaction means in plain words
Tranylcypromine and Valbenazine are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
Tranylcypromine belongs to Central Nervous System; Valbenazine belongs to Central Nervous System.
Why it happens
Opposing effects on monoamine disposition: VMAT2 inhibition versus MAO inhibition preventing monoamine breakdown.
What to do
Do not combine; select an alternative therapy. Observe the washout period recommended in the labeling when changing between agents.
