摘要
Amyl nitrite is a rapid, potent vasodilator, and tadalafil greatly enhances its hypotensive effect. Profound hypotension, collapse and cardiac ischemia can result, including with recreational inhaled nitrites.

Amyl nitrite and Tadalafil are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.

Nitrite-derived nitric oxide raises cGMP and PDE5 inhibition prevents its degradation, causing excessive additive vasodilation.

Do not use together; allow at least 48 hours after the last tadalafil dose before any nitrate. Advise patients to avoid recreational nitrites entirely.
