Summary
Amiodarone inhibits several CYP enzymes and P-glycoprotein and could modestly raise loratadine levels. Loratadine has a wide safety margin and is not known to cause meaningful QT effects at normal doses, so clinical significance is limited.
What this interaction means in plain words
Amiodarone and Loratadine have a small, usually unimportant interaction. Most people can take both; the pharmacist may still mention it so that any unexpected effect is recognised.
Amiodarone belongs to Cardiovascular; Loratadine belongs to Respiratory.
Why it happens
Inhibition of CYP2D6/CYP3A4 metabolism and P-glycoprotein efflux by amiodarone.
What to do
No dose adjustment is usually necessary. Be aware of additional drowsiness and review ECG monitoring already required for amiodarone.
