Summary
Fedratinib inhibits CYP3A4 and may raise exposure to monomethyl auristatin E (MMAE), the cytotoxic payload released from brentuximab vedotin. This could increase the risk of neutropenia and peripheral neuropathy, though the effect of a moderate inhibitor has not been well characterized.
What this interaction means in plain words
Brentuximab and Fedratinib can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Brentuximab belongs to Oncology; Fedratinib belongs to Oncology.
Why it happens
Fedratinib inhibition of CYP3A4 may reduce clearance of free MMAE.
What to do
Monitor blood counts and watch for emerging peripheral neuropathy and infection. Consider dose adjustment of brentuximab if toxicity develops.
