Summary

Lonafarnib is a CYP3A4 inhibitor and may raise bromocriptine levels, while bromocriptine as a weak CYP3A4 inhibitor may modestly raise lonafarnib exposure. Increased exposure to either drug could worsen adverse effects, including hypotension with bromocriptine and gastrointestinal toxicity or arrhythmia risk with lonafarnib.

What this interaction means in plain words

Bromocriptine and Lonafarnib can interact in a way that may be serious. Using them together is sometimes necessary, but it needs a deliberate decision by the prescriber, a plan for monitoring, and clear advice on the warning signs to watch for.

Evidence level: Low

Why it happens

Mutual CYP3A4 inhibition, with lonafarnib being a sensitive CYP3A substrate and inhibitor.
