Summary

Safinamide is a MAO-B inhibitor and dexmethylphenidate is a CNS stimulant that raises synaptic dopamine and norepinephrine. Combining them can precipitate a hypertensive crisis and hyperadrenergic or serotonergic toxicity. The labeling of both classes advises against concurrent use.

What this interaction means in plain words

Dexmethylphenidate and Safinamide are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.

Dexmethylphenidate belongs to Central Nervous System; Safinamide belongs to Central Nervous System.

Evidence level: Moderate

Why it happens

MAO inhibition reduces catecholamine breakdown while the stimulant increases catecholamine release and blocks reuptake, producing additive sympathomimetic effects.

What to do

Do not co-administer; allow a washout of at least 14 days after stopping safinamide before starting dexmethylphenidate. Choose a non-interacting alternative for ADHD.

What to ask your pharmacist
