Summary

Etrasimod is cleared by CYP2C8, CYP2C9 and CYP3A4. Teriflunomide is a weak CYP2C8 inhibitor, and in CYP2C9 poor metabolizers this could raise etrasimod exposure. Teriflunomide also adds immunosuppressive effects.

What this interaction means in plain words

Etrasimod and Teriflunomide can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.

Etrasimod belongs to Gastrointestinal Tract; Teriflunomide belongs to Central Nervous System.

Evidence level: Low

Why it happens

Weak CYP2C8 inhibition by teriflunomide may reduce etrasimod clearance, and both drugs have additive immune effects.

What to do

Avoid the combination in known CYP2C9 poor metabolizers. In other patients, monitor for etrasimod adverse effects such as bradycardia and for infections.
