For healthcare professionals — reference only. This page is a brief summary, not medical advice and not a substitute for official labeling or clinical judgement. Always verify before prescribing or dispensing.
Overview
Fluconazole can increase rosuvastatin exposure, raising risk of statin-associated myopathy/rhabdomyolysis. Effect is usually modest but clinically relevant at higher statin doses or in predisposed patients.
Mechanism of action
Fluconazole inhibits CYP2C9/2C19 (minor metabolic pathways for rosuvastatin), leading to a modest increase in rosuvastatin concentrations.
Clinical management
Use lowest effective rosuvastatin dose during fluconazole therapy; consider dose reduction if using >20–40 mg/day. Monitor for muscle pain/weakness; check CK and liver enzymes if symptomatic; hold rosuvastatin if myopathy suspected.
