Summary

Androgen deprivation with leuprolide can lengthen the QT interval, and lefamulin also prolongs QTc. Used together, the additive effect may raise the risk of torsades de pointes, especially in patients with electrolyte disturbances or heart disease.

What this interaction means in plain words

Lefamulin and Leuprolide can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.

Lefamulin belongs to Anti Bacterial; Leuprolide belongs to Endocrine.

Evidence level: Low

Why it happens

Additive pharmacodynamic QT prolongation through different mechanisms (hormonal effect on cardiac repolarization and ion channel effects of lefamulin).

What to do

Obtain a baseline ECG and correct potassium and magnesium before combining. Repeat the ECG after starting lefamulin, or choose a non-QT-prolonging antibiotic if the patient has other risk factors.

What to ask your pharmacist

  • Should I take them at different times of day?
  • Does the dose of either medicine need to change?
  • Is there anything I should avoid, such as alcohol or certain foods, while on both?
