Summary

Tipranavir is given with ritonavir, and the boosted regimen strongly inhibits CYP3A4, increasing lovastatin exposure. This raises the risk of myopathy and rhabdomyolysis.

What this interaction means in plain words

Lovastatin and Tipranavir are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.

Lovastatin belongs to Cardiovascular; Tipranavir belongs to Anti Virals.

Evidence level: Moderate

Why it happens

Net CYP3A4 inhibition by ritonavir-boosted tipranavir reduces lovastatin clearance.

What to do

Do not use together; switch to a statin less dependent on CYP3A4 such as pravastatin or rosuvastatin. Monitor for muscle symptoms.
