Summary
Tipranavir is given with ritonavir, and the combination has complex but net strong CYP3A4 inhibitory effects that can raise lurasidone concentrations. This increases the risk of dose-related adverse effects including sedation and extrapyramidal symptoms. The combination is contraindicated.
What this interaction means in plain words
Lurasidone and Tipranavir are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
Lurasidone belongs to Central Nervous System; Tipranavir belongs to Anti Virals.
Why it happens
Ritonavir-boosted tipranavir inhibits CYP3A4-mediated metabolism of lurasidone.
What to do
Avoid the combination; use an alternative antipsychotic or antiretroviral.
