Summary
Ospemifene is metabolized by CYP3A4 and CYP2C9, and CYP3A4 inhibitors raise its exposure. Ribociclib may therefore increase ospemifene levels, and ospemifene has an estrogen agonist effect on the endometrium and carries thromboembolic warnings. It is also not suited to patients with breast cancer.
What this interaction means in plain words
Ospemifene and Ribociclib can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Ospemifene belongs to Endocrine; Ribociclib belongs to Oncology.
Why it happens
Inhibition of CYP3A4-mediated ospemifene metabolism by ribociclib.
What to do
Avoid ospemifene in patients with breast cancer; if it is considered, use an alternative for vulvovaginal atrophy and monitor for adverse effects.
