Summary
Voriconazole inhibits CYP3A4 and can increase exposure to both amlodipine and atorvastatin. This may cause hypotension and peripheral edema from amlodipine, and raises the risk of myopathy or rhabdomyolysis from atorvastatin.
What this interaction means in plain words
Amlodipine-atorvastatin and Voriconazole can interact in a way that may be serious. Using them together is sometimes necessary, but it needs a deliberate decision by the prescriber, a plan for monitoring, and clear advice on the warning signs to watch for.
Amlodipine-atorvastatin belongs to Cardiovascular; Voriconazole belongs to Anti Fungals.
Why it happens
Voriconazole inhibits CYP3A4, reducing the metabolism of amlodipine and atorvastatin.
What to do
Monitor blood pressure and for muscle symptoms, and consider reducing the dose of amlodipine or atorvastatin during voriconazole therapy. Consider temporarily suspending the statin or using a statin not dependent on CYP3A4 for a short course.
