Summary
Voriconazole increases ceritinib exposure via CYP3A4 inhibition, and ceritinib may in turn modestly affect voriconazole metabolism. Both drugs prolong the QT interval, so arrhythmia risk is raised, along with ceritinib hepatic and gastrointestinal toxicity.
What this interaction means in plain words
Ceritinib and Voriconazole can interact in a way that may be serious. Using them together is sometimes necessary, but it needs a deliberate decision by the prescriber, a plan for monitoring, and clear advice on the warning signs to watch for.
Ceritinib belongs to Oncology; Voriconazole belongs to Anti Fungals.
Why it happens
CYP3A4 inhibition raising ceritinib levels, with additive QTc prolongation.
What to do
Avoid if possible; if needed, reduce ceritinib dose by about one-third and monitor ECG, electrolytes and liver tests. Return to the previous dose after voriconazole is discontinued.
What to ask your pharmacist
- Does the benefit of taking both outweigh the risk in my case?
- Do I need any blood test, ECG or other check while on both?
- What early warning signs should I watch for, and what should I do if they appear?
