Summary

Dacomitinib is a strong CYP2D6 inhibitor and vortioxetine is cleared primarily by CYP2D6. Vortioxetine exposure may therefore rise considerably, increasing nausea and other dose-related adverse effects. Serotonergic toxicity is a theoretical concern at higher levels.

What this interaction means in plain words

Dacomitinib and Vortioxetine can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.

Dacomitinib belongs to Oncology; Vortioxetine belongs to Central Nervous System.

Evidence level: Moderate

Why it happens

Inhibition of CYP2D6 by dacomitinib reduces vortioxetine clearance.

What to do

Consider reducing the vortioxetine dose by about half while dacomitinib is taken, as for other strong CYP2D6 inhibitors. Return to the original dose after dacomitinib is stopped and its inhibition has resolved.

What to ask your pharmacist

  • Should I take them at different times of day?
  • Does the dose of either medicine need to change?
  • Is there anything I should avoid, such as alcohol or certain foods, while on both?
