Summary
Fedratinib is a moderate CYP3A4 inhibitor and also a CYP3A4 substrate, so both drugs may raise each other's exposure. This may increase ribociclib QT prolongation and neutropenia and fedratinib toxicities such as GI effects and encephalopathy.
What this interaction means in plain words
Fedratinib and Ribociclib can interact in a way that may be serious. Using them together is sometimes necessary, but it needs a deliberate decision by the prescriber, a plan for monitoring, and clear advice on the warning signs to watch for.
Fedratinib belongs to Oncology; Ribociclib belongs to Oncology.
Why it happens
Mutual CYP3A4 inhibition increases exposure of both drugs.
What to do
Consider reducing ribociclib dose and monitor ECG, electrolytes and blood counts. Watch for fedratinib toxicity including neurologic symptoms and thiamine deficiency.
What to ask your pharmacist
