Summary

Glofitamab induces cytokine release that can transiently suppress CYP450 enzymes. Quinidine, a narrow therapeutic index CYP3A4 substrate, may accumulate and increase QT prolongation risk, especially around the first doses or during cytokine release syndrome.

What this interaction means in plain words

Glofitamab and Quinidine can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.

Glofitamab belongs to Oncology; Quinidine belongs to Anti Parasitic.

Evidence level: Moderate

Why it happens

Cytokine-mediated suppression of CYP3A4 activity lowers quinidine clearance.

What to do

Monitor ECG/QTc and quinidine levels during the first treatment cycle and during CRS. Reduce the quinidine dose if toxicity occurs.

What to ask your pharmacist

  • Should I take them at different times of day?
  • Does the dose of either medicine need to change?
  • Is there anything I should avoid, such as alcohol or certain foods, while on both?
