Summary
Givosiran has been shown to reduce CYP2D6 activity in some patients, and aripiprazole is cleared substantially by CYP2D6. Aripiprazole levels may therefore rise, increasing akathisia, sedation, and other dose-related effects.
What this interaction means in plain words
Aripiprazole and Givosiran can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Aripiprazole belongs to Central Nervous System; Givosiran belongs to Miscellaneous.
Why it happens
Givosiran-associated inhibition of CYP2D6 reduces the metabolism of aripiprazole.
What to do
Monitor for aripiprazole adverse effects such as akathisia and sedation, and reduce the aripiprazole dose if they occur. Consider the same dose reduction used for CYP2D6 poor metabolizers if the effect is pronounced.
What to ask your pharmacist
- Should I take them at different times of day?
- Does the dose of either medicine need to change?
- Is there anything I should avoid, such as alcohol or certain foods, while on both?

How serious is the interaction between Aripiprazole and Givosiran?
MediHelp rates it moderate, on a four-level scale (minor, moderate, major, contraindicated) based on published labels and references. What to do, in brief: Monitor for aripiprazole adverse effects such as akathisia and sedation, and reduce the aripiprazole dose if they occur. Consider the same dose reduction used for CYP2D6 poor metabolizers if the effect is pronounced.
