Summary
Calcifediol is cleared partly by CYP3A4-mediated hydroxylation, so itraconazole may increase exposure to calcifediol and its active metabolite. This may raise the likelihood of hypercalcemia.
What this interaction means in plain words
Calcifediol and Itraconazole can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Calcifediol belongs to Musculoskeletal; Itraconazole belongs to Anti Fungals.
Why it happens
Strong CYP3A4 inhibition slows catabolism of calcifediol.
What to do
Monitor serum calcium, phosphate and PTH when itraconazole is started or stopped. Adjust the calcifediol dose as needed.
What to ask your pharmacist
- Should I take them at different times of day?
- Does the dose of either medicine need to change?
- Is there anything I should avoid, such as alcohol or certain foods, while on both?
