Overview
Gastric acid suppression from omeprazole can reduce domperidone oral bioavailability, potentially decreasing antiemetic/prokinetic efficacy.
Mechanism of action
Increased gastric pH (antisecretory effect) may reduce domperidone dissolution/absorption; not a CYP3A4 interaction.
Clinical management
Administer domperidone 15–30 min before meals and avoid taking it simultaneously with omeprazole; separate by ≥2 hours and monitor clinical response. Adjust therapy if efficacy is reduced.
