Summary
Itraconazole raises elagolix concentrations by inhibiting CYP3A4, which may increase dose-related effects such as bone mineral density loss and hepatic transaminase elevation. Elagolix is a weak-to-moderate CYP3A inducer and may modestly lower itraconazole exposure.
What this interaction means in plain words
Elagolix and Itraconazole can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Elagolix belongs to Musculoskeletal; Itraconazole belongs to Anti Fungals.
Why it happens
Itraconazole inhibits CYP3A4 metabolism of elagolix, while elagolix may weakly induce CYP3A.
What to do
Avoid the 200 mg twice-daily elagolix dose with a strong CYP3A inhibitor beyond one month. Limit elagolix to 150 mg once daily and the inhibitor to a maximum of 6 months, and monitor antifungal efficacy.
What to ask your pharmacist
- Should I take them at different times of day?
- Does the dose of either medicine need to change?
- Is there anything I should avoid, such as alcohol or certain foods, while on both?
