Summary

Idelalisib is a strong CYP3A inhibitor and markedly increases exposure to CYP3A4 substrates. Eletriptan is labeled as contraindicated with strong CYP3A4 inhibitors within at least 72 hours because of the risk of higher exposure and vasoconstrictive adverse effects.

What this interaction means in plain words

Eletriptan and Idelalisib are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.

Eletriptan belongs to Central Nervous System; Idelalisib belongs to Oncology.

Evidence level: High

Why it happens

Strong CYP3A4 inhibition by idelalisib reduces eletriptan first-pass and systemic clearance.

What to do

Do not use eletriptan with idelalisib. Choose a triptan not metabolized by CYP3A4.
