Summary
Estrogens may raise tricyclic antidepressant exposure, increasing anticholinergic, sedative and cardiovascular adverse effects. Conversely, antidepressant efficacy has occasionally been reported to be reduced. The data are limited and inconsistent.
What this interaction means in plain words
Estradiol and Trimipramine have a small, usually unimportant interaction. Most people can take both; the pharmacist may still mention it so that any unexpected effect is recognised.
Estradiol belongs to Endocrine; Trimipramine belongs to Central Nervous System.
Why it happens
Estrogens may inhibit hepatic oxidative metabolism of tricyclics, but the mechanism is not firmly established.
What to do
Monitor for tricyclic adverse effects such as sedation, dry mouth and orthostasis, and for loss of antidepressant response when estrogen is started, stopped or changed. Adjust the trimipramine dose according to clinical response.
