Summary
Levoketoconazole is a strong CYP3A4 and P-glycoprotein inhibitor and is expected to markedly raise everolimus concentrations. Higher exposure increases the risk of serious adverse effects including infections, stomatitis, pneumonitis and metabolic disturbances.
What this interaction means in plain words
Everolimus and Levoketoconazole are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
Everolimus belongs to Oncology; Levoketoconazole belongs to Endocrine.
Why it happens
Strong inhibition of CYP3A4 and P-glycoprotein by levoketoconazole reduces everolimus clearance.
What to do
Avoid the combination; use an alternative to levoketoconazole or everolimus. Strong inhibitors such as ketoconazole are to be avoided with everolimus per labeling.
