Summary
Pretomanid inhibits the renal organic anion transporter OAT3 in vitro, and penicillin G is eliminated mainly by tubular secretion through this pathway. Exposure to penicillin G could therefore rise modestly, but the clinical relevance has not been established and penicillin G has a wide therapeutic margin.
What this interaction means in plain words
Penicillin g and Pretomanid have a small, usually unimportant interaction. Most people can take both; the pharmacist may still mention it so that any unexpected effect is recognised.
Penicillin g belongs to Anti Bacterial; Pretomanid belongs to Anti Bacterial.
Why it happens
Inhibition of OAT3-mediated renal tubular secretion of penicillin G by pretomanid, shown in vitro.
What to do
No routine dose change is needed. Watch for dose-related penicillin adverse effects such as neurotoxicity when high doses are given, especially in renal impairment.
