Summary
Posaconazole strongly inhibits CYP3A4, which markedly raises ergotamine levels. This can cause ergotism with severe peripheral and cerebral vasospasm and ischemia.
What this interaction means in plain words
Ergotamine and Posaconazole are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
Ergotamine belongs to Central Nervous System; Posaconazole belongs to Anti Fungals.
Why it happens
Posaconazole inhibits CYP3A4, blocking ergotamine metabolism.
What to do
Do not coadminister; use an alternative antimigraine therapy during and after posaconazole treatment. If exposure occurs, watch for ischemic symptoms and seek urgent care.
What to ask your pharmacist
