Summary
Glofitamab induces cytokine release that can transiently suppress CYP450 activity and raise concentrations of CYP3A4 substrates such as everolimus. The risk is highest after initial doses and during cytokine release syndrome.
What this interaction means in plain words
Everolimus and Glofitamab can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Everolimus belongs to Oncology; Glofitamab belongs to Oncology.
Why it happens
Cytokine-mediated suppression of CYP3A4 reduces everolimus metabolism.
What to do
Monitor for everolimus toxicity and check trough levels in the days after glofitamab dosing, especially at treatment initiation. Reduce the everolimus dose if concentrations or adverse effects increase.
What to ask your pharmacist
- Should I take them at different times of day?
- Does the dose of either medicine need to change?
- Is there anything I should avoid, such as alcohol or certain foods, while on both?
