Summary
Enzalutamide is a strong CYP3A4 inducer and may accelerate the hydroxylation and clearance of calcifediol. Serum 25-hydroxyvitamin D levels may fall and the intended effect on calcium and bone metabolism may be reduced.
What this interaction means in plain words
Calcifediol and Enzalutamide have a small, usually unimportant interaction. Most people can take both; the pharmacist may still mention it so that any unexpected effect is recognised.
Calcifediol belongs to Musculoskeletal; Enzalutamide belongs to Endocrine.
Why it happens
Enzalutamide induces CYP3A4-mediated catabolism of calcifediol, shortening its half-life.
What to do
Monitor serum 25-hydroxyvitamin D, calcium and PTH during long-term co-therapy. Increase the calcifediol dose if levels or clinical response are inadequate.
