Summary
Posaconazole is a strong CYP3A4 inhibitor and also inhibits P-glycoprotein, so it can markedly raise silodosin concentrations. Strong CYP3A4 inhibitors are contraindicated with silodosin because of the risk of orthostatic hypotension, dizziness and syncope.
What this interaction means in plain words
Posaconazole and Silodosin are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
Posaconazole belongs to Anti Fungals; Silodosin belongs to Genitourinary.
Why it happens
Inhibition of CYP3A4 and P-glycoprotein by posaconazole reduces silodosin clearance and increases its exposure.
What to do
Do not combine; choose an alternative alpha-blocker or antifungal. If antifungal therapy is unavoidable, hold silodosin for the duration of treatment.
What to ask your pharmacist
- Is there an alternative to one of these two medicines for me?
- If both are essential, who is monitoring me and how often?
- Which symptoms mean I should stop and seek help straight away?
